To study gene therapy, transfection in vivo with oncogene expression vectors requires using oxygen tension control systems to achieve greater levels of transfection. In this study, the efficacy of oxygen tension control in a rat model was compared with non-oxygen-controlled conditions. In rats, the transfection efficacies of five different oxygen tension control systems were compared after cotransfection of an avian retrovirus-based vector expressing enhanced green fluorescent protein (EGFP) and the oncogene tyrosine kinase, bcr-abl. The oxygen tension control systems evaluated included perfluorodecalin, perfluorotributylamine, carbogen breathing (95%O2-5%CO2), hyperoxia, and an equimolar mixture of perfluorodecalin and N2 gas. The control system that most efficiently mediated transfection of EGFP-positive cells was oxygen tension control with hyperoxia. This efficiency declined to a lower level, however, when the oncogene was expressed. All five oxygen tension control systems yielded comparable levels of apoptotic nuclei in the liver and spleen.
